不同浓度舒芬太尼对脂多糖诱导的外周血中白细胞介素-6和肿瘤坏死因子-α浓度的影响

The effect of different concentrations of sufentanil on interleukin-6 and tumor necrosis factor-α concentrations in peripheral blood induced by lipopolysaccharide

摘要:

目的 探讨不同浓度舒芬太尼对脂多糖(lipopolysaccharide, LPS)诱导的正常人离体外周血中IL-6和TNF-α浓度的影响。 方法 30例正常志愿者,采用完全随机设计分组法分为对照组(A组)、LPS组(B组)、舒芬太尼组(C组)、舒芬太尼和LPS组(D组),采集正常志愿者外周静脉血5 ml,收集在抗凝试管内,准备好标记过(A、B、C1、C2、C3、D1、D2、D3)的8个试管,用高精度加样器每管加入每例志愿者100 μl的静脉血。在8个试管中依次加入以下的8组试剂: A组加入生理盐水, B组加入LPS 100 mg/L, C组(C1组、C2组、C3组)分别加入舒芬太尼0.05、0.50、5.00 μg/L, D组(D1组、D2组、D3组)分别加入舒芬太尼0.05、0.50、5.00 μg/L和LPS 100 mg/L。用人IL-6和TNF-αELISA试剂盒检测处理后血样上清液中IL-6和TNF-α的浓度。 结果 A组和C组外周血中IL-6、TNF-α浓度差异无统计学意义(P>0.05);与A组和C组比较,B组和D组外周血中IL-6、TNF-α浓度显著增加(P<0.05);与B组比较,D组外周血中IL-6、TNF-α浓度显著降低(P<0.05);与D1组比较,D2组、D3组外周血中IL-6、TNF-α浓度降低(P<0.05);与D2组比较,D3组外周血中IL-6、TNF-α浓度降低(P<0.05)。 结论 LPS可促进人离体外周血中炎性因子IL-6和TNF-α浓度的增加;舒芬太尼能抑制LPS诱导的人离体外周血中IL-6和TNF-α浓度的增加,且呈浓度依赖性;舒芬太尼对正常人离体外周血中IL-6和TNF-α浓度均无影响。

Abstract:

Objective Investigating the effect of different concentration of sufentanil on IL-6 and TNF-α concentrations in human peripheral blood induced by lipopolysaccharide(LPS). Methods Every blood sample were collected from 30 healthy volunteers. Using a completely randomized design grouping method. Samples were randomized assigned in eight groups. Venous blood 5 ml from every volunteers were collected. 100 μl of the venous blood from every sample were put in eight ethylenediaminete-traacetic acid-vitro ready tubes labeled(A, B, C1, C2, C3, D1, D2, D3) by using precision pipette. Groups are: group A for the saline group(control), group B joined LPS 100 mg/L, group C includes C1, C2, C3, respectively fentanyl 0.05, 0.50, 5.00 μg/L, group D includes the D1, D2, D3 fentanyl 0.05, 0.50, 5.00 μg/L and LPS 100 mg/L, respectively. The IL-6 and TNF-α ELISA kit were used to detect supernatant concentrations of IL-6 and TNF-α. Results Group A and group C in peripheral blood IL-6 and TNF-α concentration were not significantly different(P>0.05). Plus LPS of group B and group D of IL-6, of TNF-α concentrations were significantly increased(P<0.05). Group D and group B, the peripheral blood of IL-6 and TNF-α concentrations were significantly lower(P<0.05). Group D2, D3 peripheral IL-6 and TNF-α concentration reduction are greater than the group D1(P<0.05). Group D3 peripheral blood IL-6 and TNF-α concentration were greater than group D2 reduction(P<0.05). Conclusions LPS can promote isolated human peripheral blood inflammatory cytokine IL-6 and TNF-α concentration increasing. Sufentanil inhibited LPS-induced, IL-6 and TNF-α concentration increase in a dose-dependent manner from the peripheral blood. Sufentanil had no effect on IL-6 and TNF-α concentration the peripheral blood drawn away from people.
