摘要:

背景 临床上常使用长效局部麻醉药予以镇痛,但由于个体差异,在药物浓度较低时可能对痛觉阻滞不全,药物浓度过高时则会阻滞运动功能,因此难以达到理想的镇痛效果。 目的 总结近期对QX-314研究的文献资料。 内容 QX-314是利多卡因的季胺类衍生物,呈永久正电荷,不易透过细胞膜。近年来发现QX-314可以经过瞬时感受器电位香草酸受体1(transient receptor potential vanlloid subtype 1, TRPV1)通道进入神经细胞,阻滞钠离子通道,产生长时间的镇痛作用,且对运动功能影响轻微。 趋向 QX-314的镇痛作用为局部麻醉药运用于临床提供了新的思路。

Abstract:

Background Generally, long-acting local anesthetic is used for analgesia in clinic, but low concentration may lead to inefficiency and high concentration may result in the block of motor function due to individual difference. Therefore, it does not meet the need of perfect analgesic effect. Objective To summarize the recent literature about the study of QX-314. Content The quaternary amine lidocaine derivative -QX-314 with permanent positive charge, dose not permeate the membrane passively. The recent studies showed that QX-314 could penetrate into nerve cell and blocked sodium channel through the transient receptor potential vanlloid subtype 1 (TRPV1) channel, and produced prolonged analgesia effect with little effect on the motor function. Trend The analgesic effect of QX-314 indicates a new way for the clinic use of regional anesthesia.
