背景 脊髓背角N-甲基-D-天冬氨酸(N-methyl-D-asparate, NMDA)受体的过度活化是慢性疼痛中枢敏化的关键环节之一。近年研究证明,非受体依赖的酪氨酸蛋白激酶Fyn能与NMDA受体(NMDA receptors, NR)调节亚基NR2B上的衔接蛋白相互作用,调节NR2B的酪氨酸磷酸化,进而调节NR的活性。 目的 进一步阐明慢性疼痛的发生机制,寻找新的疼痛治疗药物。 内容 对Fyn调节NR2B磷酸化的机制及其在慢性疼痛中枢敏化的重要作用进行综述。 趋向 干预Fyn调节的NR2B磷酸化有望成为慢性疼痛治疗的新靶点。

Abstract:

Background The overactivity of N-methyl-D-aspartate (NMDA) receptors in spinal cord dorsal horn plays a critical role in central sensitization of chronic pain. Recent studies indicate that the phosphorylation of NMDA receptors(NR) are tightly regulated by the Src family tyrosine kinase Fyn, through dynamic interactions with various scaffolding proteins in the NR2B subunit of NR. Objective This article aims to further elucidate the mechanism of chronic pain, and explore novel methods for pain management. Content The article focuses on the mechanism of phosphorylation of NR mediated by Fyn, and the essential role of Fyn in the central sensitization of chronic pain. Trend Prevention of the phosphorylation of NR2B-containing NR mediated by Fyn may become a new target for the treatment of chronic pain.
